Abstract

Lifestyle modification and dietary supplements are important components of the management of pulmonary hypertension. The aim of this study was to evaluate the effects of a low-dose (5 mg/kg) of Sildenafil (Viagra, Pfizer) on serum concentrations of the main vasodilatory aminotricoplast system and on the other blood-oxygenation-related molecules in patients with PAH. Samples were obtained from 108 patients with PAH who had a clinical history of hypertension, dyspnea, and/or other cardiovascular diseases. Samples were also obtained for other vasoactive substances and for other drugs and foods used to control pulmonary hypertension. The levels of blood-oxygenation-related compounds, as well as the major vasodilators and the drugs used to control the disease, were evaluated. The data were analysed using the Kruskal–Wallis test and the Wilcoxon matched paired samples test. The concentration of the vasodilators was significantly lower in patients with PAH than in the control group. In addition, the blood-oxygenation-related molecules showed significantly lower concentrations in the patients with PAH than in the patients with controls. A lower dose of Sildenafil (Viagra, Pfizer) significantly reduced the blood-oxygenation-related molecules in patients with PAH, while other drugs were not significantly affected by the drug levels. The results suggest that a low dose of Viagra (Viagra) may be useful in the management of patients with PAH. However, more research is needed to confirm the results of this study.

Author summary

We evaluated the effects of a low-dose of Sildenafil (Viagra) on the vasodilatory effects of the main blood-oxygenation-related compounds and on the other blood-oxygenation-related molecules in patients with pulmonary hypertension. A low-dose of Viagra (Viagra) significantly reduced the blood-oxygenation-related molecules in patients with PAH, while other drugs were not significantly affected by the drug levels. A lower dose of Sildenafil (Viagra) may be useful in the management of patients with PAH.
